Gene-Editing Found To Preserve Hearing In Mice And It Could Do The Same For Humans
There are around 11 million people in the UK, or one in six members of the population, who suffer with hearing loss, and nearly half of all these cases have a genetic cause.

Treatment has long been limited because scientists didn’t have the right technology at their disposal to treat the underlying problem, but a new study has found gene editing could provide the solution.

Investigator David Liu, who worked on the study, said: “[It] could make a major difference in hearing-loss patients’ quality of life.”

The work, which is among the first to apply genome editing to deafness in animals, looked at one mutation in a gene called Tmc1 – a single spelling error in this gene causes the loss of inner ear hair cells over time.

These delicate bristles detect sound and then convert this information into nerve signals to travel to the brain. The loss of these bristles causes profound deafness in both humans and mice.

Liu, said: “The conventional thinking in the field is that once you’ve lost your hair cells, it’s difficult to get them back.”

So the team looked at what would happen if they administered a single injection of the CRISPR-Cas9 genome editing cocktail to the inner ear on infant mice.

They hoped that the technology would be able to remove the mutation and stop the bristles from dying.

What is CRISPR? CRISPR-Cas9 technology is an enzyme that acts as an injectable ‘molecular scissor’ and can be used by scientists to edit DNA strands. For example, in an embryo found to have a problem, the tool can ‘snip’ out the offending DNA. It has already been used to remove hereditary heart conditions, potentially stop HIV in its tracks, and create low-fat pigs. Embryos modified by CRISPR are not permitted to carry children to term, it is only for purposes of “better understanding” human development. In the UK, the UK Human Fertilisation and Embryology Authority (HFEA) 2016 licensing, says embryos must be destroyed within 14 days.

After eight weeks, hair cells in treated ears resembled those in healthy animals, meaning it was densely packed and tufted with hairlike bundles. While the hair cells of untreated mice, in contrast, looked damaged and sparse.

Even just one shot was found to prevent progressive hearing loss in young animals that would have otherwise gone deaf. The regrowth of these hairs allowed them to hear sounds about 15 decibels lower than untreated ears.

“That’s roughly the difference between a quiet conversation and a garbage disposal,” Liu said.

Because the study was performed in mice, the implications for treating humans are still unclear, but they did say they hoped it would work in the same way in humans.

Work and pensions secretary David Gauke has vowed not to let the ‘hard left’ win arguments over Universal Credit on the social media ‘battlefield’.

The cabinet minister entered into a Twitter spat – with mixed results – after Labour MP and work and pensions select committee chair Frank Field claimed the roll-out of the controversial benefit system had left one woman with no income until after Christmas. 

Gauke said the woman had in fact already been paid her advance and had not been specially sought out after Field raised the case, citing it as an example of the new system – which replaces the six current main benefits – “working properly”.

Important to note that the payment was not as a consequence of FF’s tweet or an intervention from the centre. Just UC working normally.

But the Labour MP hit back on Wednesday afternoon, claiming the woman in question was still in need of help.

“It’s great that you’ve found and helped someone in such desperate need,” he tweeted.

“But the mum to whom I referred still hasn’t had an appointment. The public has ensured she has money for food and heating today. She’s still seeking help from DWP and I hope you’ll ensure this happens.”

During a briefing on Universal Credit roll-out hosted by the Centre for Social Justice, alongside former Conservative leader Iain Duncan Smith, Gauke told journalists: “I think it’s important that we don’t desert the battlefield.  Social media can at the moment feel almost dominated by the hard left, and I think it’s important that we engage and make the points.

“I strongly believe that we have got a really good policy that has positively transformed lives, but – and I am not for a moment suggesting Frank is part of the hard left – there is almost a sort of knee-jerk criticism, and a temptation, I think in particular with Universal Credit, that you can almost say anything critical and it goes without challenge.”

He said reports of problems with the system from Labour MPs had led to claimants becoming anxious about the process, while Duncan Smith blamed “irresponsible” reporting of the issue in the media. 

Gauke said: “We are definitely picking up within the Jobcentres that people are coming in to make their claim for universal credit more anxious than would have been the case three months ago. They are generally leaving the Jobcentre reassured.

“I do think there needs to be a responsible debate.  Of course there is legitimate debate about any major reform, and looking at case studies is a legitimate part of that.

“But I am concerned that there are vulnerable people who are scared, not by actual cases, but by some ill-informed criticism, or cases which are not as they are first portrayed. 

“We have seen that pretty consistently.  There have been many incidences of that, and it’s my job to try to bring a bit of balance to this debate.”

He said Jobcentre staff across the country had praised the policy and its “transformative” impact on claimant’s lives.

“I have met so many work coaches up and down the country who are consistently enthusiastic about the policy, and consistently telling me about how they are shouting at the television or the radio when they see the policy they are working on being – in their eyes – misrepresented,” he added.

“Yes it is big, yes it is ambitious, but it’s something that on the ground is working well.”

Gauke said take-up of advance payments – which are available as loans repayable via deductions from future Universal Credit allowances – had risen from about 34% in the spring to 62%, according to latest figures. 

He also pledged to keep the benefit’s ‘taper rate’ – the amount every pound of Universal Credit is reduced by if a claimant earns over their work allowance – under review after it was lowered from 65p initially to 63p.

“The point both Philip [Hammond] and I have made is that yes, it will be good if we could bring it down further, but clearly that is expensive and has to be viewed in light of public finances.

“But the aspiration to bring it down further is there.”

Labour has long called for the roll-out of the system to be paused while problems with its implementation are ironed out – with many MPs warning their constituents would be pushed into poverty as a result of delayed initial payments. 

“They are dealing at the end of the day with people who are often in quite vulnerable situations, and to run these kind of stories as though this were a crisis I have to tell you I think is verging on the irresponsible,” the former cabinet minister said.

“These facts should be fully checked before stories are pushed out and that also goes for the opposition, who are at the moment playing a political game with this. 

“Their aim is to make things look as bad as possible.  But frankly it is pretty cynical to try and do that, and I would suggest that people behave a little more responsibly, because people’s lives matter in this regard.”

Yes, after yet another year of sly comments, clunky put-downs and an exchange of jibes that ended with a concertgoer peeling a potato in front of Liam, it seems the duo might be able to sit down and enjoy the festive season together after all.

Liam revealed the news on Tuesday (21 December) night, when he took to Twitter to wish Noel – well, “Team NG” – a happy Christmas:

Am Dienstagmorgen wird es zunächst ruhig und kalt. Im Osten ist es klar mit mäßigem Frost und am Alpenrand fallen die Werte auf -13 Grad. Im Westen ist dagegen schon die Warmluft angekommen.

Auf Sylt bleibt es am Morgen 8 Grad warm. In den westlichen Landesteilen bildet sich sehr dichter Nebel. Am Dienstag breitet sich die Warmluft weiter aus. Dazu kommt Regen aus Norden auf.

An den Mittelgebirgen schneit es zunächst noch, aber der Schnee geht immer mehr in Regen über. Das Weihnachtstauwetter beginnt. Es kann nochmal glatt werden auf den Straßen. Im Süden und Osten hält sich noch die kalte Luft.

Ein Verkehrschaos droht

In der Nacht zum Mittwoch erreicht der Schneefall dann den Südwesten. Es schneit kräftig und es mischt sich auch zusehends Regen unter den Schnee. Am Mittwochmorgen droht im Südwesten ein Verkehrschaos. Gleichzeitig setzt massives Tauwetter in den Mittelgebirgen ein. Nur im Osten bleibt es noch kalt. Die Tiefstwerte liegen zwischen +7 Grad im Nordwesten und -10 Grad an den Alpen.

Bis zu 30 Zentimeter Neuschnee

Am Mittwoch kommt der Schneefall nach Osten voran. Es droht nochmal Eisregen in Schwaben und Bayern. Gleichzeitig muss mit leichtem Hochwasser besonders am Schwarzwald, am Rothaargebirge und am Harz gerechnet werden.

Am Donnerstag wird es regnerisch. Nur an den Alpen über 1300 Meter schneit es noch. Dafür ergiebig. Es fallen bis zu 30 Zentimeter Neuschnee. Der Freitag wird auch ungemütlich, nass und grau.

Das Wetter an Weihnachten bleibt mild. Im Süden wird es am Heiligen Abend eher sonnig, im Norden bedeckt sein. Am 1. Feiertag wird es verbreitet sonnig und am 2. Feiertag kann es nochmal über 600 Meter einzelne Schneeschauer geben. Aber dafür wird es stürmisch im Westen.

A man has died in a motorway crash following a police chase in Bradford. 

Officers began pursuing a silver Audi A4 at around 2.45am this morning (Monday) after the driver failed to stop in the city’s Heaton district, West Yorkshire Police reported. 

According to the force, the Audi crashed into a barrier on the M606 – near the M62 junction – a “short time later”.

The driver, a man in his thirties, was pronounced dead at the scene. His family have been informed. 

The road remains closed while investigations into the collision are underway. 

The incident has been reported to the Independent Police Complaints Commission (IPCC).

A spokesperson for the commission said: “We have begun an independent investigation into a fatal road traffic incident at around 3 am today (Monday) following a police pursuit in Bradford.

“The driver of an Audi vehicle died at the scene on the M606 southbound at the junction with the M62.

“After being notified by West Yorkshire Police, we have deployed investigators to the incident and the police post incident procedures.”

Noch bevor die GroKo-Sondierungen begonnen haben, zoffen sich SPD und Union schon wie die Kesselflicker.

Vor allem die Sozialdemokraten halten sich nicht mit Kritik am Verhandlungspartner nicht zurück. Neustes Beispiel: Stephan Weil.

“Mancher aus der CSU sitzt noch auf einem hohen Ross”

Die Union sollte der SPD aus Sicht des niedersächsischen Ministerpräsidenten mit mehr Respekt und Kooperationswillen begegnen.

“Ohne die SPD geht nichts. Das sollte sich auch die Union klarmachen”, sagte er der “Welt am Sonntag”.

Ein Satz lässt in dem Interview besonders aufhorchen. “Mancher, vor allem in der CSU, sitzt noch immer auf einem hohen Ross”, sagte Weil.

Das erinnert an die gegenseitigen Angriffe bei den Jamaika-Verhandlungen – und lässt nichts Gutes erahnen. 

Die SPD-Führung hatte sich am Freitag nach langem internen Ringen dafür ausgesprochen, nun doch Sondierungen mit CDU und CSU über eine Regierungsbildung aufzunehmen.

“Müssen fleißig und effizient arbeiten”

Nach einem Vorbereitungstreffen der Spitzen von Union und SPD an diesem Mittwoch sollen die Sondierungen im Januar starten.

Weil rief die Union und seine eigene Partei auf, nun “fleißig und effizient” zu arbeiten. Die Bürger seien der ewigen, fruchtlosen Gespräche überdrüssig.

Weil sagte, es müsse klar werden, wo die Reise hingeht.

“Brauchen keine Spiegelstrichdiskussion”

“Wir brauchen keine Spiegelstrich-Diskussionen, keine 300-Seiten-Papiere, sondern lange Linien”, sagte der Ministerpräsident.

Die CDU-Vorsitzende, Kanzlerin Angela Merkel, habe bei den schließlich gescheiterten Gesprächen über eine Koalition mit FDP und Grünen “Hunderte eckiger Klammern zugelassen und die Verhandlungen sind aus dem Ruder gelaufen”.

Diese Bruchlandung habe nicht nur FDP-Chef Christian Lindner zu verantworten, sondern auch Merkel.

Auch SPD-Mann Ulrich Kelber war am Freitag vor allem damit beschäftigt, die Union zu attackieren. “Kackfrech” sei die Forderung der CSU nach Grenzkontrollen in Deutschland. 

Kackfrech fordert die CSU bessere Grenzkontrollen an der Ost- und Westgrenze Deutschlands. Dabei sind derzeit massenhaft BundespolizistInnen zB aus NRW nach Bayern abkommandiert und fehlen im Westen

Doch auch die Union hat längst begonnen, die angestrebten Verhandlungen zu einem kindischen Hick-Hack zu verwandeln.

Jamaika lässt grüßen

Jens Spahn etwa erklärte unlängst, “Kompromisse mit der SPD müssen auf Basis des Regelwerks von CDU und CSU entstehen“. Im Prinzip heißt das: Die SPD hat bei den Sondierungsgesprächen die Wahl, dem Unionsprogramm zuzustimmen, oder nicht. 

Und die von der SPD ins Spiel gebrachten sozialen Projekte für eine schwarz-rote Koalition lehnen die Konservativen mit stoischer Konsequenz ab. So zuletzt die Bürgerversicherung durch CDU-Minister Hermann Gröhe.

Gin moved up from third place last year as 29% of consumers named it their favourite spirit, ahead of whisky (25%) and vodka (23%), the annual poll by the Wine and Spirit Trade Association (WSTA) found.

Sales of the spirit have doubled in value in the last six years to £1.2 billion in the 12 months to September, up from £630 million in 2011, WSTA’s market report figures show.

The equivalent of more than 8.8 million bottles of gin were sold in pubs, bars and restaurants to a value of £729 million in the same 12 months, while 38.7 million bottles were sold in shops and supermarkets.

In the 12 weeks up to September, gin sales were up 26% by volume in shops and supermarkets and up 34% by value compared to the same time last year.

HMRC figures early in the year showed that gin exports were outperforming those of British beef and soft drinks, the Press Association reported.

WSTA chief executive Miles Beale said: “The latest WSTA market report is showing yet another sparkling result for gin sales in the UK.

“The British public show no signs of growing tired of trying new gins with well over 100 brands now available on the UK market.

“It comes as no surprise that gin has climbed to the top of a poll of most popular spirit drinks.”

The spirit’s popularity has seen producers offering up an increasing range of locally-sourced botanicals, such as seaweed, rhubarb and Christmas gins with frankincense and myrrh.

A total of 45 new distilleries opened in the UK during 2016, according to HMRC, taking the total to an estimated 273, more than double 2012’s 128.

In March, the Office for National Statistics added it to its typical shopping basket of goods used to calculate inflation.

In June, ‘Brighton Gin’ was named the best vintage in the People’s Drink Awards.
